Introduction

The iLenti siRNA vector represents a third generation of lentiviral vectors derived from HIV-based lentivirus. The iLenti siRNA vector is designed with convergent Pol III promoters that drive the transcription of target siRNA from opposite directions. The subcloning of target siRNA is achieved by a simple ligation of annealed target siRNA to the Bbs I linearized iLenti vector. The linearized iLenti vector contains two different 5’ overhangs to reduce the occurrence of background self-ligation and mis-directional cloning. The iLenti siRNA vector represents a third generation of lentiviral vectors derived from HIV‐1 based lentivirus. The third generation design incorporates significant number of safety features to enhance its biosafety and minimize the possibility of generation replication competent lentivirus.The trans-acting function of Tat becomes dispensable if part of the upstream LTR in the transfer vector construct is replaced by constitutively active promoter sequences. Furthermore, the expression of rev in trans allows the production of high-titer HIV-derived vector stocks from a packaging construct which contains only gag and pol. This design makes the expression of the packaging functions conditional on complementation available only in producer cells. The resulting gene delivery system, which conserves only three of the nine genes of HIV-1 and relies on four separate transcriptional units for the production of transducing particles, offers significant advantages for its predicted biosafety.This includes the separation of genes encoding viral structures and other components for viral packaging into 3 different plasmids and genetic mutations in the viral 5’ and 3’ LTR region. The 3 plasmids used for recombinant lentiviral production contain no regions of homology to each other to prevent undesirable homologous recombination leading to the generation of replication competent viral particles. Mutations in 5’ and 3’ LTR regions render the recombinant lentivirus incapable of producing a viral genome that can be packaged once integrated into the transduced target cells.
