Advantages of siRNA-pLV Constructs:
- RNAi in lentiviral vectors can be used for transfection or viral infection.
- siRNA-pLV vectors are very stable and can be amplified in regular competent cells.
- Lentiviral vectors offer the highest, stable gene transduction efficiency.
- Choice of antibiotic resistance markers: Neo or Puro
- siRNA-pLV uses 27-29 bp oligos, which are more efficient than traditional 21mers.
- siRNA-pLV uses convergent promoters to avoid hairpin loop structures, making sequencing and plasmid growth much easier.
- siRNA-pLV saves money on oligos, as ~35 instead of 70bp oligos are used for the expression of double standed RNAi in vivo.
