Custom Avenovirus Construction
Recombinant adenoviruses are a versatile and highly efficient tool used for gene delivery and expression in mammalian cells. A number of biological features of adenovirus have made such viruses the vectors of choice for both in vitro and in vivo applications. Adenoviruses can infect a broad range of cell types with the highest efficiency and infection is not dependent on active host cell division. A second key feature is that high virus titers and high-level gene expression can be obtained in most mammalian cells.
A recombinant adenovirus containing full-length cDNA will allow high level gene expression in host cells, whereas adenovirus construct containing RNAi oligos specific to the target gene will result in efficient inhibition of its expression.
Adenovirus vector construction and virus production includes the following steps:
- Cloning gene of interegst (GOI) into pShuttle vector
- Subcloning GOI into the adenovirus genome from pShuttle vector
- Production of virus seed stock (low titer prep)
- Amplification and purification (high titer prep)
